Build Custom Enzyme List

There are two ways to build a custom enzyme list.

Import Enzymes From a File

Enzymes can be imported from a CSV file. Enzymes included in the CSV file can be either new enzymes that are not available in the system or enzymes already stored in the system. The first line of the CSV file should be headers (names of primers properties), the second line and lines after it should contain property values of a particular enzyme, and values are separated with semicolons. The fields to be included in the input file are name (enzyme name), sequence (restriction site sequence), cut (cutting positions), offset, provider, inactivation temperature, and incubation temperature.  The mandatory fields are name (enzyme name), sequence (restriction site sequence), cut (cutting positions), and offset. An example of the CSV file is shown below.

 

  1. On any sequence tab, select Action -> Import -> Enzyme

  1. Enter the name of the custom enzyme list. Then, press “Create”.

 

  1. In the import enzymes window, either click “Choose file” to select the file that contains enzyme information or drag and drop the import file to the window.

  1. After enzymes are imported from the file, the newly imported list is added under Custom Lists in the enzyme panel. The enzymes are displayed on the scene and are visible in the enzyme panel if the custom list is selected.

 

 

Create Enzyme List Manually

The user can create a preferred enzyme list from enzymes stored in the system and add commonly used/favorite enzymes to it.

 

  1. In the Enzymes panel, expand the enzyme list dropdown and press “+ CREATE NEW LIST”.

 

  1. Enter the name of the custom enzyme list. Then, press “Create”.

 

  1. If “All Enzymes” is selected in the enzyme list dropdown, all default enzymes will be displayed. Pick the enzymes that will be added to the list by checking the box before the enzyme name. If the enzyme is not on the default list but in a custom list, expand the enzyme list dropdown and select the custom list that contains the enzyme(s). After the custom enzyme list is displayed, check the preferred enzyme.

  1. After all enzymes are selected, press “ADD TO LIST” to create the new custom list. The new list will be added under “Custom lists”
